RecA K72R filament formation defects reveal an oligomeric RecA species involved in filament extension.
Using an ensemble approach, we demonstrate that an oligomeric RecA species is required for the extension phase of RecA filament formation. The RecA K72R mutant protein can bind but not hydrolyze ATP or dATP. When mixed with other RecA variants, RecA K72R causes a drop in the rate of ATP hydrolysis and has been used to study disassembly of hydrolysis-proficient RecA protein filaments. RecA K72R ...
متن کاملFocal adhesions are hotspots for keratin filament precursor formation
Recent studies showed that keratin filament (KF) formation originates primarily from sites close to the actin-rich cell cortex. To further characterize these sites, we performed multicolor fluorescence imaging of living cells and found drastically increased KF assembly in regions of elevated actin turnover, i.e., in lamellipodia. Abundant KF precursors (KFPs) appeared within these areas at the ...
متن کاملGenes for intermediate filament proteins
The increase in specific cell types represents one hallmark of metazoan evolution. It is paralleled by the acquisition of multigene families, which often encode proteins of similar structure but distinct function. One such family is represented by the intermediate filament protein (IF) family. Its members form part of the cytoskeleton of most metazoan cells. Vertebrate IF are organised into fiv...
متن کاملOPTIMUM DESIGN OF FILAMENT–WOUND LAMINATED CONICAL SHELLS FOR BUCKLING USING THE PENALTY FUNCTION
Optimum laminate configuration for minimum weight of filament–wound laminated conical shells subject to buckling load constraint is investigated. In the case of a laminated conical shell the thickness and the ply orientation (the design variables) are functions of the shell coordinates, influencing both the buckling load and its weight. These effects complicate the optimization problem consider...
متن کاملFilament ring formation in the dimorphic yeast Candida albicans
Stationary phase cultures of Candida albicans inoculated into fresh medium at 37 degrees C synchronously from buds at pH 4.5 and mycelia at pH 6.5. During bud formation, a filament ring forms just under the plasma membrane at the mother cell-bud junction at roughly the time of evagination. A filament ring also forms in mycelium-forming cells, but it appears later than in a budding cell and it i...
